Supplementary figure 1. Hierarchical clustering of genes that show differential expression in the liver, adrenal, kidney, brain, testis, lung and heart tissues of wild type mice upon T1317 LXR agonist treatment compared with LXR alpha-/- LXR beta-/- mice -
Wild type mice and LXR alpha-/- LXR beta-/- mice were fed with either the T1317 LXR agonist or vehicle alone (four mice in each group) for one week and gene expression profiling was performed on RNA extracted from the liver (A), adrenals (B), kidneys (C), brain (D), testis (E), lung (F) and heart (G). Cluster analyses were based on 1039, 287, 234, 150, 169, 336, and 979 genes, respectively, that showed differential expression between non-treated tissues (WC1,2,3,4 (liver WC1,2,4)) and LXR agonist T1317 treated (WT5,6,7,9 (liver WT5,7,9)) in wild type mice (P<0.01). As comparison, the same gene information from LXR alpha-/- LXR beta-/- mice for non-treated tissues (KC11,12,13,14 (liver KC11,12,14)) and for agonist-treated tissue, (KT15,16,17,18 (liver KT17,168,19)) was aligned. Rows represent individual genes, and columns represent unique samples. Each cell in the matrix represents the expression level of a gene feature in an individual tissue sample. The red and green colour bars reflect high and low expression levels respectively, while black indicates equivalent expression level. The magnitude of the log-transformed ratio is reflected by the degree of colour saturation.
Supplementary figure 2. Hierarchical clustering of genes that show differential expression in the liver and adrenal tissues of wild type mice versus LXR alpha-/- LXR beta-/- mice. -
Wild type mice and LXR alpha-/- LXR beta-/- mice were fed vehicle alone for one week and gene expression profiling was performed on RNA extracted from the liver (A) and adrenals (B). Cluster analyses were based on 694 and 366 genes in the liver and adrenal, respectively, that showed differential expression between wild type mice (WC1,2,3,4 (liver WC1,2,4)) and LXR alpha-/- LXR beta-/- mice (KC11,12,13,14 (liver KC11,12,14)) (P<0.01).
